擠出原理

Extrusion Principle

樣品經(jīng)過水化形成脂質體后，一般會通過高壓均質或者超聲的方式對樣品進行初步均質處理，處理后樣品粒徑和分布都會有所提高，但往往還不能達到所需要的理想要求。此時，將均質后樣品由下圖中進料口進入，通過高壓氮氣將樣品從濾膜中擠出，擠出后的樣品粒徑和分布都會進一步提高。

After treatment, the particle size and distribution of the sample will be improved, but it can not meet the desired requirements. At this time, the homogenized sample enters through the feed port in the figure below, and the sample is extruded from the filter membrane by high-pressure nitrogen. The particle size and distribution of the extruded sample will be further improved.

支撐板

Support plate

支撐板和濾板主要起到支撐濾膜的作用。由于磷脂有一定的相變溫度，所以在擠出之前需要對擠出器夾套通入恒溫水進行預熱至一定溫度（如70℃），且在使用過程中需保持在此溫度。通常的脂質體品種擠出之后粒徑能達到 100nm以下，PDI 能控制在 0.1 以下。
